An experimental study on bone formation of fibrin/DFBP complex implanted in the peritoneal muscle fascia of the rat



Abstract
Various forms of bone graft materials have been used for reconstruction of the congenital malformations, acquired deformities after tumor surgery trauma, or physiologic bone resorption following absence of teeth in the oral and maxillofacial
region. Van
Meekren was the first one to report bone grafting. Senn transplanted demineralized bovine bone to for the clinical purpose.
In 1960s, Urist reported osteoinduction in the demineralized bone embedded in the extraskeletal sites. Thereafter Reddi, Huggins, Sharawy, Glowacki, Kaban, Mullikan, and others others reported the experimental and clinical reports of potentiality
of the
demineralized bone.
In the recent clinical practice, there are many cases using dimineralized freeze-dried bone powder (DFBP) with various forms of occlusive barriers or adhesives as a carrier, were used for the bone regeneration.
The purpose of this experiment is to find out whether fibrin clot can be used as a carrier for DFBP.
We used saline-rehydrated DBP in control group rats and saline-reconstituted DFBP mixed with fibrin in experimental group and sacrifice the rats at postoperative 3 days, 1 week, 2 weeks, 4 weeks, and 6 weeks.
We stained the demineralized samples with HE staining and the undemineralized samples with Villanueva staining and the samples was observed under the light microscope.
@ES The results were as follows.
@EN 1. The amount of the early mesenchymal tissue ingrowth at the third day specimen of the experimental was similar to that of 1st week control group.
The blood cells were found in the specimen at the postoperative third day in both of the experimental groups and the control groups.
Osteoid matrix surrounding the graft materials were observed from the postoperative 4th week in both of the experimental groups and the control groups.
Bone remodelling was observed from the postoperative 4th week in both of the experimental groups and the control groups.
